ПОЛІМОРФІЗМ ЯДЕРЦЕОРГАНІЗУЮЧИХ РАЙОНІВ ХРОМОСОМ БУЙВОЛА РІЧКОВОГО (BUBALUS BUBALIS L.)

У статті викладені результати досліджень ознаки «число ядерцеорганізуючих районів (ЯОР) [nucleolus organizer regions (NOR)] у хромосомах» буйвола річкового (Bubalus bubalis L). У ядерцеорганізуючих районах хромосом наявна рибосомальна РНК (рРНК), асоційована з аргентофільними білками ядерець, які забезпечують синтез білка в соматичних клітинах і впливають на процеси росту й розвитку організму тварин і людини. Інтенсивність фарбування районів ядерцевих організаторів відображає потенційні можливості функціонального стану клітин у сільськогосподарських тварин. Число, розміри і хромосомна локалізація зафарбованих сріблом районів організаторів ядерця є видоспецифічними ознаками.

ЯОР вивчали на препаратах хромосом, які готували за стандартною методикою з лімфоцитів периферійної крові 10 дорослих тварин, які утримуються в господарстві монастиря «Свято-Покровська Голосіївська Пустинь» — ТОВ «Голосіїво» (с. Гоголів Броварського р-ну Київської обл.). Фарбування препаратів хромосом з метою виявлення районів ядерцевих організаторів проводили 50 %-ним розчином азотнокислого срібла згідно з рекомендацями Committee for standardized karyotype of Bubalus bubalis, 1994. ЯОР виявляли на теломерах відповідних хромосом як темні цятки.

У результаті досліджень було виявлено активні райони ядерцевих організаторів у шести хромосомах каріотипу досліджених буйволів — 3, 4, 6, 21, 23, 24. Встановили індивідуальну мінливість тварин за числом ЯОР — у метафазних пластинках виявлено від 1 до 12 на клітину, із середнім числом 2,82 у 207 клітинах. Найбільшою є частка метафазних пластинок із двома ЯОР (93 із 208), більш як вдвічі меншою є частка метафаз із трьома ЯОР, по 12% — із одним та чотирма ЯОР. Показано, що Ag-метод фарбування може успішно застосовуватись в цитогенетичних дослідженнях тварин як метод оцінки функціонування генів рРНК для оцінки метаболітичної активності організму, швидкої ідентифікації аберацій хромосом, зокрема транслокацій, в які залучені хромосоми з ядерцеорганізуючими районами. Перспективним є дослідження поліморфізму ЯОР сільськогосподарських тварин на молекулярному рівні і співставлення його із показниками їх продуктивності.
